Function: - signal-transducing molecule - signal transducing subunit of several cytokine receptors - receptor systems for IL6, LIF, OSM, CNTF, IL11, CTF1 & BSF3 can utilize gp130 for initiating signal transduction - binds to IL6/IL6R (alpha chain) complex, resulting in formation of high-affinity IL6 binding sites, & transduces the signal - does not bind IL6 - may have a role in embryonic development (putative). - forms heterodimers composed of LIPR & IL6ST (type 1 OSM receptor) - also forms heterodimers composed of OSMR & IL6ST (type 2 OSM receptor) - phosphorylation of Ser-782 down-regulates cell surface expression Structure: - the WSXWS motif appears to be necessary for proper protein folding, efficient intracellular transport & cell-surface receptor binding - the box 1 motif is required for JAK interaction &/or activation - belongs to the type 1 cytokine receptor family, type 2 subfamily - contains 5 fibronectin F3 modules - contains 1 Ig-like C2-type domains (immunoglobulin-like) Compartment: - isoform 1: cell membrane - isoform 2: secreted Alternative splicing: named isoforms=2 Expression: - found in all the tissues & cell lines examined - expression not restricted to IL6 responsive cells Pathology: - isoform 2 is an autoantigen found in rheumatoid arthritis (RA) but it is not specific to patients with RA Note: - GP130 also maps to 130 kD leucine-rich protein
