WW domain-containing transcription regulator protein 1 (Transcriptional coactivator with PDZ-binding motif, WWTR1, TAZ)

Function: - transcriptional coactivator - binds to SLC9A3R2 via PDZ motif at plasma membrane - binds to YWHAZ in vivo & in vitro t via phosphoserine-binding motif RSHSSP Structure: - PDZ-binding motif a) essential for stimulated gene transcription b) localizes the protein into both punctate nuclear foci & plasma membrane-associated complexes - contains 1 WW domain - binds to transcription factors via its WW domain Compartment: - nucleus, cytoplasm - concentrates along specific portions of the plasma membrane, & accumulates in punctate nuclear bodies - phosphorylation inhibits transcriptional coactivation via YWHAZ-mediated nuclear export - when phosphorylated, WWTR1 is retained in cytoplasm by YWHAZ Expression: - highly expressed in kidney, heart, placenta, lung
