WAS/WASL-interacting protein family member 1; protein PRPL-2; Wiskott-Aldrich syndrome protein-interacting protein; WASP-interacting protein (WIPF1, WASPIP, WIP)

Function: - may have direct activity on the actin cytoskeleton - induces actin polymerization & redistribution - role with NCK1 & GRB2 in recruitment & activation of WASL - may have role in regulating subcellular localization of WASL, resulting in the disassembly of stress fibers in favor of filopodia formation - role in intracellular motility of vaccinia virus as an adapter for recruiting WASL to vaccinia virus - binds to WAS, profilin & actin - binds to WASL Structure: - binds to WAS within the N-terminal region 170, at a site distinct from the CDC42-binding site - belongs to the verprolin family - contains 1 WH2 domain Compartment: - cytoplasmic vesicle (putative) - cytoplasm, cytoskeleton (putative) - vesicle surfaces & along actin tails - co-localized with actin stress fibers - when co-expressed with WASL, no longer associated with actin filaments but accumulated in perinuclear & cortical areas like WASL - recruited to PIP5K-induced vesicle surfaces in the absence of functional WASL (putative) Alternative splicing: named isoforms=4 Expression: - highly expressed in peripheral blood mononuclear cells, spleen, placenta, small intestine, colon & thymus - lower expression in ovary, heart, brain, lung, liver, skeletal muscle, kidney, pancreas, prostate & testis
