vinculin; metavinculin (VCL)
Function:
- role in cell adhesion
- may play roles in cell morphology & locomotion
- vinculin binds to alpha-actinin, paxillin, tensin, actin filaments & phospholipids
- binding to actin occurs via the + end of F-actin
- binding of vinculin to alpha-actinin & to talin links F-actin to plasma membrane integrins while capping F-actin
- phosphorylated; on Ser, Thr & Tyr
- phosphorylation on Tyr-1133 in activated platelets affects head-tail interactions & cell spreading but has no effect on actin binding nor on localization to focal adhesion plaques
- exhibits self-association properties
- interacts with NRAP & SORBS1 (putative)
- interacts with TLN1
- interacts with SYNM
Structure:
- aceylated; mainly by myristic acid, but also small amount of palmitic acid (putative)
- belongs to the vinculin/alpha-catenin family
Compartment:
- cytoplasm, cytoskeleton
- cell junction, adherens junction
- cell membrane; peripheral membrane protein cytoplasmic side
- cytoplasmic face of adhesion plaques
Alternative splicing: named isoforms=3
Expression: Metavinculin is muscle-specific
Pathology:
- defects in VCL are associated with dilated cardiomyopathy type 1W
Laboratory:
- vinculin Ab in serum
