transcription factor NF-E4 (NFE4)
Function:
- part of the SSP complex (stage selector protein complex)
- interacts with HDAC1 & PCAF
- acetylation at Lys-43 prolongs the protein half-life by preventing ubiquitin-mediated degradation & diminishes interaction between NF-E4 & HDAC1, potentially maximizing activating ability of the factor
- ubiquitination leads to its degradation by the proteasome
- acetylation at Lys-43 prevents ubiquitination
- isoform 2 acts as a repressor of gamma-globin gene expression by preventing NFE2 & RNA polymerase 2 recruitment to the gamma-globin gene promoter . isoform 2 interacts with TFCP2
Compartment: nucleus (probable)
Alternative initiation: named isoforms=2
Expression:
- specifically expressed in fetal liver, cord blood & bone marrow
- also expressed in the K562 & HEL cell lines, which constitutively express the fetal globin genes
