Function:
- plays a role of transcription factor
- binds to recognition signal sequences (Rss heptamer) for somatic recombination of immunoglobulin & T-cell receptor gene segments
- also binds to the kappa-B motif of genes such as S100A4
- role in cell progression & differentiation
- involvement of HIVEP3 in cell growth is strengthened by the fact that its down-regulation promotes cell cycle progression with ultimate formation of multinucleated giant cells
- strongly inhibits TNF-alpha-induced NF-kappa-B activation
- interferes with nuclear factor NF-kappa-B by several mechanisms:
a) as transcription factor
b) by competing for Kappa-B motif
c) by repressing transcription in the nucleus
d) through non transcriptional process
e) by inhibiting nuclear translocation of RELA by association with TRAF2, an adapter molecule in the tumor necrosis factor signaling, which blocks formation of IKK complex
- interaction with TRAF proteins inhibits both NF-Kappa-B mediated & c-Jun N-terminal kinase/JNK mediated responses, including apoptosis & proinflammatory cytokine gene expression
- positively regulates expression of IL2 in T-cells
- role in adult bone formation
- interacts with TRAF1 & TRAF2 as well as with JUN
- forms multimeric complex with RUNX2 & E3 ubiquitin ligase WWP1
- phosphorylated on threonine & serine residues
Structure:
- the ZAS2 domain binds DNA as dimers, tetramers, & multiple of tetramers & readily forms highly ordered DNA-protein structures (putative)
- contains 5 C2H2-type Zn+2-fingers

Alternative splicing: named isoforms=2 Additional isoforms can be generated by alternative splicing or polyadenylation. HIVEP3L transcript may lack exon 7 leading to a premature codon stop
