transcriptional activator protein Pur-beta (purine-rich element-binding protein B, PURB)
Function:
1) binds repeated elements in single-stranded DNA, i.e. purine-rich single strand of the PUR element located upstream of the MYC gene
2) role in control of vascular smooth muscle alpha-actin gene transcription as repressor in myoblasts & fibroblasts
3) role in transcriptional & translational regulation of alpha-MHC expression in cardiac myocytes by binding to the purine-rich negative regulatory element
4) modulates constitutive liver galectin-3 gene transcription by binding to its promoter
5) role in dendritic transport of a subset of mRNAs
Structure:
- homodimer
- heterodimer with PURA
- heterotrimer with PURA & YBX1/Y-box protein 1
- belongs to the PUR DNA-binding protein family
Compartment: nucleus
Pathology:
- expressed in myocardium of heart failure patients
- defects in PURB may be a cause of progression of myelodysplastic syndrome towards acute myelogenous leukemia
