Function: 1) innate immune response to microbial agents 2) mediates detection of bacterial flagellins 3) acts via MyD88 & TRAF6 a) NF-kappa-B activation b) cytokine secretion c) inflammatory response 4) binds MyD88 via their respective TIR domains Structure: - belongs to the Toll-like receptor family - contains 15 LRR repeats (leucine-rich repeats) - contains 1 TIR domain Compartment: membrane Expression: - expressed in ovary, peripheral blood leukocytes, especially monocytes > CD11c+ immature dendritic cells, prostate, testis Polymorphism: - genetic variation in TLR5 is associated with resistance to systemic lupus erythematosus type 1 - individuals with a stop codon polymorphism in position 392 are unable to mediate flagellin signaling a) expressed in dominant fashion b) associated with susceptibility to pneumonia caused by Legionella pneumophila Pharmacology: - TLR5 agonist CBLB502 may be useful as radioprotective agent during radiation therapy for cancer treatment
