Function: - cytokine that regulates megakaryocyte development - acts at a late stage of megakaryocyte development - may be the major physiological regulator of circulating platelets - ligand for Mpl receptor Structure: - two-domain structure with an erythropoietin-like N-terminal & a Ser/Pro/Thr-rich C-terminal - belongs to the EPO/TPO family - at least two forms, 25 kDa & 31 kDa Compartment: secreted Alternative splicing: named isoforms=3 Pathology: - defects in THPO are a cause of essential thrombocythemia Pharmacology: - avatrombopag (Doptelet) is a thrombopoietin receptor agonist that stimulates platelet production [5] Laboratory: - thrombopoietin in plasma Comparative biology: - cDNAs for human, dog & mouse predict proteins of 353, 352 & 356 amino acids respectively.
