thrombin receptor; proteinase-activated receptor 1; PAR-1; coagulation factor 2 receptor (F2R, CF2R, PAR1, TR)

Function: - high affinity receptor for activated thrombin - coupled to G proteins that stimulate phosphoinositide hydrolysis (Gi protein & Gp protein) - may play a role in platelets activation & in vascular development - thrombin specifically cleaves its receptor in an extracellular domain leading to self activation - proteolytic cleavage generates a new N-terminus that functions as a tethered ligand - phosphorylated; probably mediating desensitization prior to the uncoupling & internalization of the receptor Structure: belongs to the G-protein coupled receptor 1 family Compartment: cell membrane; multi-pass membrane protein Expression: platelets & vascular endothelial cells
