Function: - involved in translocation of long-chain fatty acids across the plasma membrane - hormone-regulated in a tissue-specific manner - in adipocytes, insulin induces a rapid translocation of SLC27A1 from intracellular compartments to the plasma membrane (not so in myocytes); this occurs in parallel with increased LFCA uptake - may act directly as a bona fide transporter, or in a cytoplasmic or membrane-associated multimeric protein complex to trap fatty acids for accumulation - role in regulating available LFCA substrates from exogenous sources in tissues undergoing high levels of beta-oxidation or triglyceride synthesis - may be involved in regulation of cholesterol metabolism - acyl-CoA ligase activity for a) long-chain fatty acids b) very long-chain fatty acids Structure: - self-associates, may function as an homodimer - belongs to the ATP-dependent AMP-binding enzyme family Compartment: - plasma membrane & intracellular membranes, at least in adipocytes - predominantly cytoplasmic in myocytes Expression: - expressed in muscle & adipose tissue > small intestine > liver Pathology: - FATP1-mediated fatty acid uptake may be implicated in: a) insulin resistance b) fatty acid metabolism & homeostasis c) obesity
