Function: - mediates Na+-independent transport of organic anions such as estrone-3-sulfat - mediates transport of prostaglandins (PGE1 & PGE2, thyroxine (T4), deltorphin II, BQ-123 & vasopressin, but not DPDPE (a derivative of enkephalin lacking an N-terminal Tyr), estrone-3-sulfate, taurocholate, digoxin nor DHEAS Kinetic parameters: - KM=101 nM for PGE1 (isoform 1) - KM=218 nM for PGE1 (isoform 2) - KM=219 nM for PGE2 (isoform 1) - KM=371 nM for PGE2 (isoform 2) Structure: - belongs to the organo anion transporter (TC 2.A.60) family - contains 1 Kazal-like domain Compartment: - membrane - in choroid plexus epithelium - isoform 1 is localized at the basolateral membrane - isoform 2 is localized at the apical membrane, as well as in the subapical intracellular vesicular compartments Alternative splicing: named isoforms=4 Expression: - ubiquitous - highly expressed in spleen & leukocytes - generally, expression of isoform 1 is higher than that of isoform 2 - isoform 2 is particularly abundant in testis & brain - in testis - isoform 1 is - detected in spermatogonia at different stages - absent from Sertoli cells - isoform 2 is - present in both spermatogonia & Sertoli cells (at protein level) - expressed in choroid plexus epithelium - differential expression of both isoforms is observed in various brain regions: - choroid plexus epithelium - isoform 1 - very abundant in the gray matter of frontal cortex - not associated with neuronal cell bodies - not detected in the white matter - isoform 2 - associated with neuronal bodies & axons in both gray matter & white matter of the frontal cortex
