Function: 1) histone methyltransferase 2) specifically methylates Lys-4 of histone H3, inducing di- & tri-methylation, but not monomethylation 3) role in transcriptional activation as component of an RNA polymerase complex 4) histone methyltransferase activity stimulated by HSPCA 5) interacts with HSPCA & HELZ, which functions as bridge between SMYD3 & RNA polymerase 2 6) binds DNA containing 5'-CCCTCC-3' or 5'-GAGGGG-3' sequences S-adenosyl-L-methionine + histone L-lysine S-adenosyl-L-homocysteine + histone 6-N-methyl-L-lysine Structure: - contains 1 MYND-type zinc finger Compartment: - cytoplasm, nucleus - mainly cytoplasmic when cells are arrested at G0/G1 - accumulates in nucleus at S phase & G2/M Alternative splicing: named isoforms=2 Expression: expressed in skeletal muscle, testis Pathology: - over-expressed in a majority of: a) colorectal carcinoma b) hepatocellular carcinomas
