Function: - may play role in hematopoietic differentiation - positive regulator of erythroid differentiation - efficient DNA binding requires dimerization with another bHLH protein - forms heterodimers with TCF3 - binds to the LIM domain containing protein LMO2 & to DRG1 - can assemble in a complex with LDB1 & LMO2. - component of a TAL1 complex composed at least of CBFA2T3, LDB1, TAL1 & TCF3 (putative) - phosphorylated on serine residues. phosphorylation of Ser-122 - strongly stimulated by hypoxia (putative) - ubiquitinated; subsequent to hypoxia-dependent phosphorylation of Ser-122; ubiquitination targets the protein for rapid degradation via the ubiquitin system; process may be characteristic of microvascular endothelial cells, since not observed in large vessel endothelial cells (putative) Structure: - helix-loop-helix domain is necessary & sufficient for interaction with DRG1 - contains 1 basic helix-loop-helix (bHLH) domain Compartment: nucleus (putative) Alternative splicing: - named isoforms=3 - splicing pattern is cell-lineage dependent Expression: leukemic stem cell Pathology: - implicated in the genesis of hematopoietic malignancies - chromosomal translocation t(1;14)(p32;q11) involving TAL1 with T-cell receptor alpha chain (TCRA) gene may be a cause of acute lymphoblastic leukemia
