replication factor-A (RFA/RPA)

Function: - single-stranded DNA-binding protein - role in DNA replication, homologous recombination & nucleotide excision repair [4] - increases processivity of DNA polymerase alpha & is required for efficient DNA polymerase delta activity - phosphorylation of RPA2 by p34cdc2 (CDK1) may activate RPA activity & DNA replication; hyperphosphorylation may inactivate RPA, halt replication & enhanc DNA repair. - DNA-binding activity may reside exclusively in RPA1 Structure: - heterotrimer a) RPA1 (70 kD) b) RPA2 (30/32 kD) c) RPA3 (14 kD) Pathology: - required for SV40 replication in vitro
