Function: - cation- & diacylglycerol (DAG)-regulated guanine nucleotide exchange factor activating Ras through exchange of bound GDP for GTP - may function in mast cells differentiation Structure: - phorbol-ester/DAG-type Zn+2 finger mediates the binding & the functional activation by DAG - belongs to the RASGRP family - contains 1 EF-hand domain - contains 1 N-terminal Ras-GEF domain - contains 1 phorbol-ester/DAG-type Zn+2 finger - contains 1 Ras-GEF domain Compartment: - cytoplasm, cell membrane - recruited to membranes upon activation by DAG Alternative splicing: named isoforms=4 Expression: - expressed by mast cells & their progenitors (at protein level) - specifically expressed in mononuclear leukocytes - highly expressed in myeloid cells compared to lymphoid cells - detected in heart, skeletal muscle, spleen, liver, placenta & lung - not detected in brain - expressed early during development - detected in all fetal tissues tested
