TRIO & F-actin-binding protein; protein tara; trio-associated repeat on actin (TRIOBP, KIAA1662, TARA, HRIHFB2122)
Function:
- may regulate actin cytoskeletal organization, cell spreading & cell contraction by directly binding & stabilizing filamentous F-actin
- localized formation of TARA & TRIO complexes coordinates the amount of F-actin present in stress fibers
- may also serve as a linker protein to recruit proteins required for F-actin formation & turnover
- ubiquitinated by HECTD3, leading to its degradation by the proteasome
- binds to TRIO & F-actin
- may also interact with myosin II
- interacts with HECTD3
Structure:
- contains at least 2 actin-binding sites per coiled-coil dimer
- contains 1 PH domain
Compartment:
- nucleus, cytoplasm, cytoskeleton
- localized to F-actin in a periodic pattern
Alternative splicing: named isoforms=5
Expression:
- widely expressed
- highly expressed in heart & placenta
- isoform 3
- expressed in fetal brain, retina & cochlea
- not detectable in the other tissues
Pathology:
- defects in TRIOBP are the cause of deafness autosomal recessive type 28
