Function: - E3-type small ubiquitin-like modifier (SUMO) ligase - stabilizes interaction between UBE2I & substrate - SUMO-tethering factor - role in transcriptional coregulation in cellular pathways a) STAT pathway b) steroid hormone signaling pathway - effects of transcriptional coregulation, transactivation or silencing may depend upon context - sumoylated (putative) - binds SUMO1 & UBE2I - interacts with AR, BCL11A, GFI1, HMGA2, IRF1, MITF, NCOA2, in addition to STAT3, after treatment with IL6, CNTF or OSM interacts with STAT5 after PRL stimulation - interacts with PLAG1 - interacts with ZFHX3 Structure: - the LXXLL motif is a transcriptional coregulator signature - belongs to the PIAS family - contains 1 PINIT domain - contains 1 SAP domain - contains 1 SP-RING-type Zn+2 finger Compartment: nuclear speckle (putative) Expression: - widely expressed - induced by dihydrotestosterone in prostate cancer cells
