protein C in plasma (protein C assay)
Specimen:
1) plasma (citrate)
2) stable at room temperature for 8 hours
3) stable at -20 degrees C for 1 month
Reference values:
1) 70-140% of normal reference activity
2) 2.82-5.65 ug/mL
Methods:
1) functional* clotting assay based on anticoagulant activity
2) rocket immunoelectrophoresis
3) enzyme-linked immunosorbant assays (ELISA)
* NOT useful if patient is anticoagulated
Increases:
-> pharmaceutical agents:
a) chemical interferences
-> heparin, warfarin
b) in vivo effects
-> oral contraceptives, stanozolol
Decreases:
1) pharmaceutical agents
-> chemical interferences
- high factor VIII activity
2) clinical disorders: hypercoagulability
a) congenital deficiency
- type 1: low functional & antigenic levels
- type 2: low functional & normal antigenic levels
b) acquired deficiency
- liver disease
- acute thrombosis [1]
- warfarin therapy
- protein-losing enteropathy [1]
