protein ajuba (AJUBA, JUB)
Function:
- component of cellular adhesives complexes
- contributes to cell fate determination & regulates cell proliferation & differentiation
- regulation of actin cytoskeleton dynamics & cell migration
- role in linking &/or strengthening of epithelia cell-cell junctions in part by linking adhesive receptors to the actin cytoskeleton
- role in signal transduction from cell adhesion sites to the nucleus
- role in regulation of the kinase activity of AURKA/Aurora-A for mitotic commitment
- component of the IL-1 signaling pathway modulating IL-1-induced NF-kappa-B activation by influencing the assembly & activity of PRKCZ/SQSTM1/TRAF6 multiprotein signaling complex
- can function as adapter molecule in formation of large multiprotein complexes that form on DNA & that influence transcription
- interacts with AURKA/Aurora-A during mitosis & both proteins are phosphorylated as they form a complex.
- interacts with CTNNA1/alpha-catenin & with F-actin
- interacts with LATS2 during mitosis & this complex regulates organization of the spindle apparatus through recruitment of gamma-tubulin to the centrosome
- interacts with GRB2 & PI51A (putative)
- forms a complex with SQSTM1, PRKCZ & TRAF6
- interacts with GLT1 (putative)
- phosphorylated by LATS2 during mitosis
- phosphorylated by AURKA
Structure:
- LIM region interacts with CTNNA1
- preLIM region binds directly actin filaments
- LIM-2 & LIM-3 domains mediate the interaction with the N-terminal region of AURKA
- association between LATS2 & JUB requires 2nd LIM domain of JUB
- belongs to the zyxin/ajuba family
- contains 3 LIM Zn+2-binding domains
Compartment:
- cytoplasm, cell membrane, cell junction, nucleus, centrosome
- shuttles between the cytoplasm & the nucleus
- localizes on centrosomes during G2-M phase
- preferentially co-localizes with cadherin-adhesive complexes at sites of cell-cell contacts
Notes: 'Ajuba' means 'curiosity' in Urdu, an Indian dialect
