Function: - RAC guanine nucleotide exchange factor (GEF), which activates Rac proteins by exchanging bound GDP for free GTP - synergistically activated by PI-3,4,5-triphosphate & G-beta-gamma - may function downstream of heterotrimeric G proteins in neutrophils - interacts preferentially with RAC2 (putative) - interacts with RAC1 Structure: - contains 1 DEP domain - contains 1 DH (DBL-homology) domain - contains 1 PDZ domain (DHR domain) - contains 2 PH domains Compartment: - cytoplasm, cell membrane - mainly cytosolic - some amount is associated with the plasma membrane Alternative splicing: named isoforms=3 Expression: - mainly expressed in peripheral blood leukocytes & brain - expressed at intermediate level in spleen & lymph nodes - weakly expressed in other tissues
