PDZ & LIM domain protein 7 (LIM mineralization protein, LMP, protein enigma, PDLIM7,ENIGMA)

Function: 1) scaffold on which the coordinated assembly of proteins can occur 2) role as adapter, localizes LIM-binding proteins to actin filaments of both skeletal muscle & nonmuscle tissues (via its PDZ domain) 3) role in bone formation a) embryonic flat bones, mandible & cranium b) endochondral bone formation, embryonic long bones 4) role in bone fracture repair 5) role in BMP6 signaling pathway 6) binds via its LIM zinc-binding 3 domain (LIM 3) to endocytic codes of INSR, but not those of IGF1R, LDLR, TFRC, or EGFR 7) interacts with various PKC isoforms through the LIM zinc- binding domains 8) binds to RET via its LIM zinc-binding domain 2 (LIM 2) (phosphorylation-independent) 9) component of a complex with SHC & the RET dimer 10) interacts with TPM2, TBX4, TBX5 - LIM zinc-binding 2 (LIM 2) domain interacts with TBX4 - LIM zinc-binding 3 (LIM 3) domain interacts with TBX5 11) anchored to cell periphery via its N-terminal PDZ domain Structure: - contains 3 LIM zinc-binding domains - contains 1 PDZ domain (DHR domain) Compartment: - cytoplasm, cytoskeleton - colocalizes with RET to cell periphery & in some cytoskeletal components - colocalizes with TPM2 near the Z line in muscle - co-localizes with TBX4 & TBX5 to actin filaments Alternative splicing: named isoforms=6 Expression: - isoforms 1 & 2 are e ubiquitous - isoform 2 predominates in skeletal muscle, - isoform 1 is more abundant in lung, spleen, leukocytes, fetal liver
