Function: - growth factor - role in regulation of embryonic development, cell proliferation, cell migration, cell survival & chemotaxis - potent mitogen for cells of mesenchymal origin - required for - normal lung alveolar septum formation during embryogenesis - normal development of the GI tract - normal development of Leydig cells - spermatogenesis - normal oligodendrocyte development - normal myelination in the spinal cord & cerebellum - role in wound healing - may provide neuronal signal for glial differentiation in CNS - signaling is modulated by the formation of heterodimers with PDGFB (putative) - homodimer; antiparallel disulfide-linked dimer - heterodimer with PDGFB; antiparallel disulfide-linked dimer - the PDGFA homodimer interacts with PDGFRA homodimers, & with heterodimers formed by PDGFRA & PDGFRB - the heterodimer composed of PDGFA & PDGFB interacts with PDGFRA homodimers, & with heterodimers formed by PDGFRA & PDGFRB - interacts with CSPG4 Structure: - the long form contains a basic insert which acts as a cell retention signal - belongs to the PDGF/VEGF growth factor family Compartment: - secreted - released by platelets upon wounding Alternative splicing: named isoforms=2; long, short Expression: platelets
