FK506-binding protein 5 (51 kD FK506-binding protein, 54 kD progesterone receptor-associated immunophilin, FKBP54, FF1 antigen, HSP90-binding immunophilin, androgen-regulated protein 6, FKBP5)

Function: - interacts with functionally mature hetero-oligomeric progesterone receptor complexes along with HSP90 & TEBP - part of a heteromultimeric cytoplasmic complex with HSP90, HSP70 & steroid receptors - dissociates from the complex when NR3C1 binds glucocorticoid - inhibited by FK506 but not cyclosporin - phosphorylated upon DNA damage, probably by ATM or ATR peptidylproline (omega=180) peptidylproline (omega=0) Structure: - contains 2 PPIase FKBP-type domains - contains 3 TPR repeats Compartment: cytoplasm. nucleus Expression: - widely expressed - enriched in testis compared to other tissues - induced by androgen Pathology: - elevated expression of FKBP5 protein promotes inflammation [2] - aging, stress, & depression increase expression of FKBP5 [2] - increased expression of FKBP5 increases expression of NF-kappa B [2]
