P2Y5 purinoceptor; oleoyl-L-alpha-lysophosphatidic acid receptor; P2Y purinoceptor 5; P2Y5; purinergic receptor 5; RB intron encoded G-protein coupled receptor (P2RY5, LPAR6)
Function:
- binds to oleoyl-L-alpha-lysophosphatidic acid (LPA)
- intracellular cAMP is involved in the receptor activation
- important for the maintenance of hair growth & texture
Structure: belongs to the G-protein coupled receptor 1 family
Compartment: cell membrane; multi-pass membrane protein
Expression:
- expressed ubiquitously, including in skin & hair follicle cells
- detected in both Henle's & Huxley's layers of the inner root sheath of the hair follicle & in suprabasal layers of the epidermis (at protein level)
- expressed at low levels in peripheral blood leukocytes
- markedly up-regulated in promyelocytic HL60 cells induced to differentiate along the monocyte/macrophage pathway
- not detectable in undifferentiated HL60 cells
- only low levels after the induction of differentiation along the granulocytic pathway
Pathology:
- defects in P2RY5 are a cause of:
a) autosomal recessive woolly hair
b) localized autosomal recessive hypotrichosis type 3
Genetics:
- gene is nested within intron 17 of the retinoblastoma gene
