FK506-binding protein 2 (13 kD FK506-binding protein, FKBP2)

Function: - associates with ryanodine receptor (RYR-2) in cardiac muscle sarcoplasmic reticulum - may play a physiological role in excitation-contraction coupling in cardiac muscle - there are four molecules of FKBP12.6 per heart muscle RYR - PPIases accelerate the folding of proteins - catalyzes the cis-trans isomerization of proline imidic peptide bonds in oligopeptides inhibited by both FK506 & rapamycin peptidylproline (omega=180) peptidylproline (omega=0) Structure: - belongs to the FKBP-type PPIase family, FKBP1 subfamily - contains 1 PPIase FKBP-type domain Compartment: cytoplasm (putative) Alternative splicing: named isoforms=2 Expression: - ubiquitous for both isoform - highest levels in brain & thymus Pathology: - may contribute to the immunosuppressive & toxic effects of FK506 & rapamycin.
