NSFL1 cofactor p47 (p97 cofactor p47, NSFL1C)
Function:
1) decreased ATPase activity of VCP
2) necessary for fragmentation of Golgi stacks during mitosis & for VCP-mediated reassembly of Golgi stacks after mitosis
3) role in VCP-mediated formation of transitional endoplasmic reticulum (putative)
4) component of ternary complex containing STX5A, NSFL1C & VCP
5) forms homotrimer that binds to one end of VCP homohexamer
- the complex binds to membranes enriched in phosphatidylethanolamine-containing lipids
- interaction with VCIP135 leads to dissociation of the complex via ATP hydrolysis by VCP
6) binds ubiquitin & mono-ubiquitinated proteins via its N-terminal UBA-like domain when bound to VCP
7) phosphorylated during mitosis
- phosphorylation inhibits interaction with Golgi membranes & is required for fragmentation of the Golgi stacks during mitosis
Structure: contains 1 UBX domain
Compartment:
- nucleus, Golgi
- predominantly nuclear in interphase
- bound to the axial elements of sex chromosomes in pachytene spermatocytes
- small proportion is associated with Golgi stacks
Alternative splicing: named isoforms=3
