Function: 1) sorting receptor in the Golgi 2) clearance receptor on the cell surface 3) protein transport from the Golgi apparatus to lysosomes (M6PR-independent) 4) protein transport from Golgi to endosomes 5) promotes neuronal apoptosis by mediating endocytosis of proapoptotic precursor forms of BDNF (proBDNF) & NGFB (proNGFB) 6) receptor for neurotensin 7) scavenges extracellular LPL - mineralization of extracellular matrix during osteogenic differentiation - induced during osteoblast differentiation 8) formation of specialized storage vesicles containing glucose transporter SLC2A4/GLUT4 in adipocytes - vesicles provide a stable pool of SLC2A4 & confer increased responsiveness to insulin 9) transport from endoplasmic reticulum to Golgi (putative) 10) interacts with LPL, SLC2A4, GGA1, GGA2, LRPAP1/RAP, GM2A, PSAP 11) forms complex with NGFR which binds to precursor forms of NGFB (proNGFB) & BDNF (proBDNF) Structure: - N-terminal propeptide may facilitate precursor transport within Golgi - N-terminal propeptide is cleaved by furin - intrachain binding of the N-terminal propeptide & extracellular domain may inhibit premature ligand binding - extracellular domain may be shed following protease cleavage - contains 9 BNR repeats Compartment: - membrane, endoplasmic reticulum, endosomes, Golgi stack, lysosomes, nucleus, plasma membrane (small fraction), SLC2A4/GLUT4 storage vesicles in adipocytes - localization to plasma membrane in adipocytes may be enhanced by insulin Expression: - expressed in brain, spinal cord, heart, skeletal muscle, thyroid, placenta, testis > lymphoid organs, kidney, colon, liver
