Function: 1) provides a protective, lubricating barrier against particles & infectious agents at mucosal surfaces 2) binds to MSLN 3) binding to MSLN mediates heterotypic cell adhesion Structure: - heavily N- & O-glycosylated - contains 2 ANK repeats - contains 14 LRR repeats (leucine-rich repeats) - contains 56 SEA domains Compartment: - cell membrane, secreted protein - may be secreted following the phosphorylation of intracellular C-terminus which induces proteolytic cleavage & liberation of extracellular domain Expression: - expressed in corneal & conjunctival epithelia Polymorphism: number of repeats is highly polymorphic Pathology: 1) heterotypic cell adhesion may contribute to the metastasis of ovarian cancer to the peritoneum by initiating cell attachment to the mesothelial epithelium via binding to MSLN 2) overexpressed in ovarian carcinomas & ovarian low malignant potential (LMP) tumors as compared to expression in normal ovarian tissue & ovarian adenomas Laboratory: - cancer antigen CA125 in tissue - cancer antigen CA125 in body fluid - cancer antigen CA125 in serum
