Function: 1) endocytotic lectin receptor (Ca+2-dependent) 2) internalizes glycosylated ligands via clathrin-mediated endocytosis 3) role in plasminogen activation, regulating extracellular PLAUR/PLAU, & thus regulation of protease activity at the cell surface (putative) 4) uptake & degradation of extracellular collagen matrices co-operating with matrix metalloproteinases (putative) 5) interacts with COL1A1, PLAUR, PLAU, collagen-5 6) phosphorylated Structure: - N-glycosylated - contains 8 C-type lectin domains a) C-type lectin domains 3 to 8 are not required for Ca+2-dependent binding of mannose, fucose & N-acetylglucosamine b) C-type lectin domain 2 is responsible for sugar-binding (Ca+2-dependent) - contains 1 fibronectin type-II domain - fibronectin type-II domain mediates collagen-binding - contains 1 ricin B-type lectin domain - ricin B-type lectin domain contacts with the 2nd C-type lectin domain (putative) Compartment: membrane Expression: - ubiquitous - low expression in brain, placenta, lung, kidney, pancreas, spleen, thymus & colon - expressed in endothelial cells, fibroblasts & macrophages - highly expressed in fetal lung & kidney Pathology: - may play role in cancer progression & other chronic tissue destructive diseases acting on collagen turnover
