lysyl oxidase or protein lysine-6-oxidase

Function: - cross-linking of extracellular matrix proteins - responsible for the post-translational oxidative deamination of Lys in precursors to fibrous collagen & elastin - direct role in tumor suppression - the Lys-Tyr quinone cross-link (LTQ) is generated by condensation of the epsilon-amino group of a Lys with a topaquinone produced by oxidation of Tyr - propeptide plays a role in directing the deposition of enzyme to elastic fibers, via interaction with tropoelastin (putative) peptidyl-L-lysyl-peptide + O2 + H2O peptidyl-allysyl-peptide + NH3 + H2O2 Cofactor: Copper Structure: - belongs to the lysyl oxidase family - contains 1 lysine tyrosylquinone (putative) Compartment: secreted, extracellular space Expression: - heart, placenta, skeletal muscle, kidney, lung & pancreas Pathology: - defects in LOX may be a cause of autosomal recessive cutis laxa type 1
