lymphotoxin-alpha; LT-alpha; TNF-beta; tumor necrosis factor ligand superfamily member 1 (LTA, TNFB, TNFSF1)
Function:
1) anti-inflammatory properties
2) stimulates fibrosis
3) generally inhibitory for B-cells
- inhibits immunoglobulin secretion
4) generally inhibitory for T-helper cells
5) generally inhibitory for cytotoxic T-cells
6) inhibits cytotoxicity of mononuclear phagocytes & NK cells
7) inhibits allergic responses
a) inhibits IgE synthesis
b) inhibtis activity of IL-4
c) inhibits mast cell proliferation
8) homotrimer binds to TNFRSF1A/TNFR1, TNFRSF1B/TNFBR & TNFRSF14/HVEM
9) heterotrimer with LTB binds to TNFRSF3/LTBR
10) cytotoxic for a wide range of tumor cells in vitro & in vivo
11) heterotrimer of either two LTB & one LTA subunits or (less prevalent) two LTA & one LTB subunits
Structure:
- homotrimer
- belongs to the tumor necrosis factor family
Compartment:
- secreted, membrane
- homotrimer is secreted; heterotrimer is membrane-associated
Expression:
- expressed & secreted by lymphocytes
Polymorphism:
- polymorphisms in lymphotoxin-alpha (LTA) accounts, in part, for susceptibility to leprosy linked to chromosome 6p21.3
- polymorphisms in lymphotoxin-alpha (LTA) are a cause of susceptibility psoriatic arthritis
