Tradenames: Zaditor, Zaditen, Alaway, Claritin Eye, Zyrtec Itchy Eye Drops Indications: -> allergic conjunctivitis Dosage: 1 drop in affected eye(s) BID Ophthalmic solution: 0.025%, 1 mL, 5 mL 0.345 mg ketotifen fumarate (0.25 mg ketotifen)/mL Contains: glycerol, benzalkonium chloride 0.01% pH: 4.4-5.8, osmolality 210-300 mOsm/kg Storage: 4-25 C Pharmacokinetics: -> systemic concentrations below detection (20 pg/mL) after topical administration Adverse effects: 1) ocular: -> allergic reactions, conjunctivitis, eye pain, keratitis, mydriasis, photophobia, rash 2) non-ocular -> flu-like syndrome, pharyngitis Toxicity: -> no serious toxicity after ingestion of 20 mg of ketotifen fumarate (1.725 mg/5 mL bottle) Mechanism of action: 1) non-competitive histamine H1 receptor antagonist 2) mast cell stabilizer 3) decreased chemotaxis & activation of eosinophils
