Kelch-like protein 12 (CUL3-interacting protein 1, KLHL12, C3IP1)

Function: 1) serves as a substrate-specific adapter for the CUL3-based ubiquitin-protein E3 ligase complex 2) negatively regulates Wnt signaling pathway via targeted ubiquitination & subsequent proteolysis of DVL3 3) component of ubiquitin-protein E3 ligase complex which includes CUL3 & KLHL12 - complex interacts with DVL3 upon activation of Wnt signaling pathway by WNT3A Structure: - BTB domain is required for interaction with CUL3 contains 1 BTB domain (POZ domain contains 6 Kelch repeats Expression: - expressed in testis & at lower levels in submandibular salivary gland
