interleukin-1 receptor antagonist protein; IL-1ra; IL-1RN; IRAP; IL1 inhibitor; ICIL-1RA (IL1RN, IL1F3, IL1RA)
Function:
- inhibits the activity of IL-1 by binding to its receptor
- has no IL-1 like activity
- has anti-inflammatory properties
- secreted normally during inflammatory response
- inhibits LPS-mediated stimulated production of:
a) IL-1 beta
b) TNF
c) IL-6
d) GM-CSF
- inhibits synthesis of IgE
Compartment:
- isoform 1: secreted
- isoforms 2, 3 & 4: cytoplasm
Alternative splicing: named isoforms=4
Expression:
- the intracellular form of IL1RN is predominantly expressed in epithelial cells
Pharmacology:
- available under the name anakinra (Kineret from Amgen)
- used for treatment of moderately to severely active rheumatoid arthritis
Pathology:
- SNP in IL-1RN associated with increased severity of osteoarthritis [8]
