Function: 1) transcriptional regulator - promotes transcription repression - promotes transcription activation in differentiated myotubes 2) binds to double- & single-stranded DNA sequences 3) binds to transcription suppressor CATR sequence of the COX5B promoter 4) binds with high affinity to RNA molecules that contain AU-rich elements found within the 3'-UTR of many proto-oncogenes & cytokine mRNAs 5) binds both to nuclear & cytoplasmic poly(A) mRNAs 6) poly(G) & poly(A), but not to poly(U) or poly(C) RNA homopolymers 7) binds to the 5'-ACUAGC-3' RNA consensus sequence 8) interacts with ZNF148, TNPO1 Structure: - dimethylation of Arg-408 is probably of asymmetric type - contains 2 RRM domain (RNA recognition motif) Compartment: - nucleus, cytoplasm - shuttles between nucleus & cytoplasm, TNPO1-dependent Alternative splicing: named isoforms=3 Expression: - expressed in heart, brain, placenta, lung, liver, skeletal muscle, kidney, pancreas, spleen, thymus, prostate, testis, ovary, small intestine, colon & leukocytes - up-regulated by 12-O-tetradecanoylphorbol 13-acetate in macrophages & retinoic acid in granulocytes - down-regulated by interleukin-4 Pathology: - expressed in myeloid leukemia, gastric adenocarcinoma, cervical carcinoma, hepatoma, fibrosarcoma, colon adenocarcinoma, epidermoid carcinoma, osteosarcoma, urinary bladder carcinoma cells
