Function: - receptor for short chain fatty acids - activity is coupled to the formation of inositol 1,4,5-trisphosphate, intracellular Ca2+ mobilization, the activation of ERK 1/ERK2 & inhibition of intracellular cAMP accumulation - coupled exclusively to the pertussis toxin-sensitive Gi-alpha &/or Go-alpha - the rank order of affinity for fatty acids is: propionate = pentanoate = butyrate > acetate > formate Structure: - belongs to the G-protein coupled receptor 1 family Compartment: -cell membrane; multi-pass membrane protein Expression: - highest expression in adipose tissue - lower expression across all tissues tested Polymorphism: - sequence differences between FFAR3 & GPR42 are due to polymorphisms - the consensus sequences within the coding regions are identical for the two genes
