GMP synthase [glutamine-hydrolyzing]; GMP synthetase; glutamine amidotransferase (GMPS)

Function: - role in the de novo synthesis of guanine nucleotides - essential for DNA & RNA synthesis - essential for providing GTP - purine metabolism; GMP biosynthesis; GMP from XMP (L-Gln route): step 1/1 ATP + xanthosine 5'-phosphate + L-glutamine + H2O = AMP + diphosphate + GMP + L-glutamate Structure: - homodimer (putative) - contains 1 glutamine amidotransferase type-1 domain - contains 1 GMP-binding domain Compartment: cytoplasm Pathology: - chromosomal translocation t(3,11)(q25,q23) involving GMPS with MLL is found in acute myeloid leukemias
