Function: 1) localized scaffold for the assembly of a multiprotein signaling complex 2) mediator of trafficking of its binding partners in neurons 3) interacts with EPHA7, EPHB2, KIF5A, KIF5B, KIF5C, GRIA2, GRIA3, GRASP1, PPFIA1, PPFIA4, FRAS1, PTPRF, EFNB1, EFNB3, liprin-alpha(s) 4) may form homomultimers or heteromultimers with GRIP2 5) forms ternary complex with GRIA2 and CSPG4 6) binds to the C-terminus of ephrin B1 through a PDZ domain 7) GRIP1 also binds to the SVKI motif in the C-terminal region of the GluR1 & GluR2 subunits of the AMPA receptor Structure: - PDZ 6 mediates interaction with the PDZ recognition motif of EFNB1 & EPHB2 & with the C-terminus of PPFIA1 & PPFIA4 PDZ 4 & PDZ 5 mediate interaction with the C-terminus of GRIA2 & GRIA3 - PDZ 4, PDZ 5 & PDZ 6 mediate homomultimers - PDZ 7 mediates interaction with PDZ domain of GRASP1 - PDZ 7 domain binds CSPG4 - PDZ 6 mediates interaction with the C-terminus of liprins-alpha - PDZ 1, PDZ 2 & PDZ 3 mediate interaction with the PDZ-binding motif of FRAS1 (putative) - PDZ 4 & PDZ 5 mediate interaction with PRLHR - contains 7 PDZ domain (DHR domain)s Compartment: - cytoplasmic & membrane-associated with vesicles - peri-Golgi complexes, endoplasmic reticulum - enriched in post-synaptic membrane & post-synaptic densities Alternative splicing: named isoforms=2
