Function: - ligand for tyrosine-protein kinase receptors AXL, TYRO3 & MER - implicated in cell growth & survival, cell-cell adhesion & cell migration - role in thrombosis by amplifying platelet aggregation & secretion - role in endothelial cell survival during acidification by preventing apoptosis - role in optimal cytokine signaling during human natural killer cell development - role in hepatic regeneration - role in gonadotropin-releasing hormone neuron survival & migration - isoform 1 is proteolytically processed after secretion to yield a N-terminal 36 kD protein & a C-terminal 50 kD protein including the laminin G-like domains which activates AXL - gamma-carboxyglutamate residues are formed by vitamin K dependent carboxylation - these residues are essential for binding of Ca+2 (putative) - heterodimer & heterotetramer with AXL Structure: - contains 4 EGF-like domains - contains 1 Gla (gamma-carboxy-glutamate) domain - contains 2 laminin G-like domains - Gas6 has significant homology with protein S Compartment: secreted Alternative splicing: named isoforms=3 Expression: - plasma - isoform 1 & isoform 2 are widely expressed - isoform 1 is the predominant form in spleen
