Fas ligand; tumor necrosis factor ligand superfamily member 6; Fas antigen ligand; CD95L protein; apoptosis antigen ligand; APTL; CD178 (FASLG, APT1LG1, FASL, TNFSF6)

Function: - cytokine that binds to FAS (CD95, TNFRSF6), a receptor that transduces the apoptotic signal into cells - may be involved in cytotoxic T-cell mediated apoptosis & in T-cell development - FasL-mediated cytotoxicity & perforin-mediated target cell lysis appear to be the 2 major effector pathways in T-cell cytotoxicity [3] - FasL-mediated apoptosis may have a role in the induction of peripheral tolerance, in the antigen-stimulated suicide of mature T-cells, or both - binding to the decoy receptor TNFRSF6B/DcR3 modulates its effects - the soluble form derives from the membrane form by proteolytic processing Structure: - homotrimer (probable) - N-glycosylated - belongs to the tumor necrosis factor family Compartment: - cell membrane, secreted - may be released into the extracellular fluid, probably by cleavage from the cell surface Alternative splicing: named isoforms=2 Expression: - expressed on the surface of activated T cells & in testis Pathology: - defects in FASL are the cause of autoimmune lymphoproliferative syndrome type 1B
