Function: 1) transcriptional activator 2) role in regulating epithelial cell differentiation & proliferation (putative) 3) repressor for a specific subset of ETS/AP-1-responsive genes 4) modulcates nuclear response to mitogen-activated protein kinase signaling cascades 5) binds to DNA sequences containing the consensus nucleotide core sequence GGAA 6) role in regulation of TNFRSF10B/DR5 expression through Ets- binding sequences on the TNFRSF10B/DR5 promoter 7) role in development & carcinogenesis by acting as a tumor suppressor gene or anti-oncogene Structure: - belongs to the ETS family - contains 1 ETS DNA-binding domain - contains 1 PNT domain (pointed domain) PNT domain acts as a transcriptional activator Compartment: nucleus Alternative splicing: named isoforms=2 Expression: - expressed exclusively in tissues with epithelial cells - expressed in salivary gland, mammary gland, prostate, lung > kidney, colon - not expressed in heart, brain, placenta, liver, skeletal muscle, spleen, thymus, testis, ovary, small intestine or peripheral blood leukocytes - induced by interleukin-1 beta & TNF-alpha in bronchial smooth muscle cells & fibroblasts - induced by interleukin-10 & IFN-gamma in monocyte-derived dendritic cells
