Function: 1) involved in base excision repair of DNA damaged by oxidation or by mutagenic agents 2) DNA glycosylase activity towards 5-hydroxyuracil & other oxidized derivatives of cytosine 3) preference for mismatched double stranded DNA (DNA bubbles) 4) low or no DNA glycosylase activity towards: thymine glycol, 2-hydroxyadenine, hypoxanthine, 8-oxoguanine 5) AP (apurinic/apyrimidinic) lyase activity - introduces nicks in DNA 6) cleaves the DNA backbone by beta-delta elimination to generate a single-strand break at the site of removed base 7) removes damaged bases from DNA, leaving an abasic site. 8) acetylation of Lys-49 leads to loss of DNA nicking activity 9) binds EP300 Structure: - the Zn+2-finger domain is important for DNA binding - belongs to the FPG family - contains 1 FPG-type Zn+2 finger Compartment: nucleus Alternative splicing: named isoforms=3 Expression: - expressed in testis, skeletal muscle, heart, brain, placenta, lung, pancreas, kidney, liver
