Endonuclease 8-like 1 (Nei-like 1, DNA glycosylase/AP lyase Neil1, DNA-[apurinic or apyrimidinic site] lyase Neil1, NEH1, FPG1, NEIL1)

Function: 1) role in base excision repair of DNA damaged by oxidation or by mutagenic agents 2) acts as DNA glycosylase that recognizes & removes damaged DNA bases - leaves an abasic site - preference for oxidized pyrimidines, i.e. thymine glycol, formamidopyrimidine, 5-hydroxyuracil - marginal activity towards 8-oxoguanine 3) AP (apurinic/apyrimidinic) lyase activity & introduces nicks in DNA 4) cleaves DNA backbone by beta-delta elimination to generate a single-strand break at the site of the removed base with both 3'- & 5'-phosphates 5) DNA glycosylase/lyase activity towards mismatched uracil & thymine, in particular in U:C & T:C mismatches Structure: - belongs to the FPG family Compartment: nucleus Alternative splicing: named isoforms=2 Expression: - ubiquitous - up-regulated during S-phase
