Function: - actin-binding protein - role in regulation of neuronal process formation - role in differentiation of neural crest cells - substrate-specific adapter of an E3 ubiquitin-protein ligase complex which mediates ubiquitination & subsequent proteasomal degradation of target proteins - protein modification; protein ubiquitination - ubiquitinated & probably targeted for proteasome- independent degradation - binds to RB1 - hypophosphorylated RB1 associates with ENC1 during neuronal differentiation - hyperphosphorylated RB1 associates with ENC1 in undifferentiating cells - part of a complex that contains CUL3, RBX1 & ENC1 Structure: - contains 1 BTB/POZ domain - contains 6 kelch repeats Compartment: - nucleus matrix. cytoplasm, cytoskeleton Expression: - detected in fetal brain tissue - moderate expression in fetal heart, lung & kidney - highly expressed in adult brain, particularly high in hippocampus & amygdala - highly expressed in adult spinal cord - detected in adult pancreas - up-regulated upon neuronal differentiation - induced by TP53/p53 Pathology: - may be down-regulated in neuroblastoma tumors
