DNA replication factor Cdt1; double parked homolog; DUP (CDT1)

Function: 1) cooperates with CDC6 to promote loading of mini-chromosome maintenance complex onto chromatin to form pre-replication complex necessary to initiate DNA replication 2) binds DNA (sequence-, strand-, & conformation-independent) 3) potential oncogene protein 4) interaction with geminin (GMNN) inhibits both binding of MCM complex to origins of replication & DNA-binding activity which may prevent inappropriate replication origin firing 5) ubiquitinated, leading to its subsequent degradation 6) interaction with GMNN protects it against ubiquitination 7) phosphorylation by cyclin A-dependent kinases resulting in binding of CDT1 to F-box protein SKP2 & subsequent degradation 8) binding to GMNN is not affected by phosphorylation Structure: - belongs to the Cdt1 family Compartment: - nucleus Expression: 1) present during G1 phase & early S phase of cell cycle 2) degraded during the late S phase, G2 phase, & M phase
