DNA damage up-regulated protein; DDUP (CTBP1-DT)

Function: - promotes DNA damage repair through both homologous recombination repair (HRR) & post-replication repair (PRR) mechanisms - enhances retention of DNA damage response protein RAD18 at sites of DNA damage - this allows for HRR via association of RAD18 with RAD51C & for PRR via RAD18-mediated promotion of PCNA monoubiquitination - phosphorylated in an ATR-dependent manner - phosphorylation is required for interaction with H2AX & RAD18 & for DDUP-mediated DNA damage repair - interacts with DNA damage response proteins ATR, H2AX, PCNA, RAD18 & RAD51C - forms a complex with H2AX & RAD18 following DDUP phosphorylation Compartment: - nucleus, chromosome - forms DNA damage-induced foci Expression: induced by DNA damage Pharmacology: - treatment of cells with ATR inhibitor Berzosertib abolishes the interaction of DDUP with H2AX, RAD18 & RAD51C & inhibits formation of DDUP foci, leading to hypersensitivity of cancer cells to DNA-damaging chemotherapeutics
