DNA-binding death effector domain-containing protein 2 (FADD-like anti-apoptotic molecule 3, DED-containing protein FLAME-3, DEDD2, FLAME3, PSEC0004)

Function: - role in death receptor-induced apoptosis - may target CASP8 & CASP10 to the nucleus - may regulate degradation of intermediate filaments during apoptosis - role in general transcription machinery in the nucleus - regulates activity of GTF3C3 (putative) - interacts with CASP8, CASP10, GTF3C3 - homodimerizes & heterodimerizes with DEDD Structure: - contains 1 DED damain (death effector domain) interactions with CASP8 & CASP10 are mediated by the DED domain Compartment: - nucleus - accumulates in subnuclear structures resembling nucleoli Alternative splicing: named isoforms=2 Expression: - expressed in most tissues - high levels found in liver, kidney, heart, ovary, spleen, testes, skeletal muscle, peripheral blood leukocytes - expression absent or low in colon, small intestine Pathology: - expression is variable in tumor cell lines
