Function: - required for CDT1 proteolysis in response to DNA damage through the CUL4-DDB1 E3 ubiquitin-protein ligase - role in cell cycle regulation of DNA replication (putative) - substrate receptor for CUL4-DDB1 E3 ubiquitin-protein ligase complex - probable part of a cullin-RING E3 protein ligase complex - interacts with CUL4B & DDB1 - phosphorylated upon DNA damage, probably by ATM or ATR Structure: - belongs to the WD repeat cdt2 family. - contains 7 WD repeats Compartment: - nucleus, nucleoplasmic side, cytoplasm - nuclear matrix-associated - translocates from interphase nucleus to the metaphase cytoplasm during mitosis Alternative splicing: named isoforms=2 Expression: - expressed in placenta & testis - very low expression in skeletal muscle - detected in all hematopoietic tissues examined - highest expression in thymus & bone marrow - low level detected in the spleen & lymph node - barely detectable level in the peripheral leukocyte - retinoic acid down-regulates expression in NT2 cells - expressed in all fetal tissues examined, including brain, lung, liver, & kidney Pathology: - role in cell proliferation of NT2 embryonal carcinoma cells
