dedicator of cytokinesis protein2 (DOCK2, KIAA0209)
Function:
1) cytoskeletal rearrangements for lymphocyte migration in response of chemokines
2) guanine nucleotide exchange factor (GEF), exchanges bound GDP for free GTP (putative)
- activates RAC1 & RAC2 small GTPases guanine
3) IL2 transcriptional activation via activation of RAC2
4) interacts with RAC1, RAC2, CRKL, VAV, CD3Z
Structure:
- the DHR-2 domain may mediate the GEF activity
- belongs to the DOCK family
- contains 1 DHR-1 (CZH-1) domain
- contains 1 DHR-2 (CZH-2) domain
- contains 1 SH3 domain
Compartment:
- endomembrane, peripheral membrane
- cytoplasm, cytoskeleton
- colocalizes with F-actin
Alternative splicing:
- named isoforms=2
- splicing donor & acceptor sites between exon 6 & exon 7 are not canonical
Expression:
- specifically expressed in hematopoietic cells, peripheral blood leukocytes > thymus, spleen > small intestine, colon
